检测原理：多采用双抗体夹心 ELISA 法。将抗人 CK - HMW 抗体包被在酶标板上，加入标准品或待测样本，样本中的 CK - HMW 与包被抗体结合，洗去未结合物质。随后加入生物素化的抗人 CK - HMW 抗体，它会与已结合的 CK - HMW 相结合，再加入辣根过氧化物酶标记的亲和素，形成抗体 - 抗原 - 生物素化抗体 - 酶标亲和素复合物。洗涤去除未结合成分后，加入显色底物 TMB，在辣根过氧化物酶催化下，TMB 显蓝色，加入终止液后变为黄色。通过酶标仪在 450nm 波长处测量吸光度，与标准曲线比较，得出样本中 CK - HMW 的浓度。

质量控制

· 绘制标准曲线：使用标准品绘制准确的标准曲线，标准品的浓度应覆盖样本中目标物的可能浓度范围。在绘制标准曲线时，要确保标准品的稀释准确，每个浓度至少设置 2 - 3 个复孔。通过标准曲线可以对样本中的目标物进行定量分析，标准曲线的线性关系和拟合度直接影响定量结果的准确性。

· 重复检测：对重要样本或检测结果有疑问的样本，进行重复检测。通过多次检测可以减少偶然误差，提高检测结果的可靠性。同时，计算多次检测结果的平均值和标准差，评估检测结果的重复性。
