检测原理：

酶联免疫吸附测定（Enzyme-Linked Immunosorbent Assay，简称ELISA）是一种基于抗原-抗体反应的生物化学分析技术，广泛应用于检测蛋白质、抗体、激素、药物等生物分子。其检测原理主要基于以下几个关键步骤：

1. 抗原-抗体特异性结合ELISA的核心是利用抗原与抗体之间的特异性结合。抗原和抗体之间通过互补的结构相互识别并结合，这种结合具有高度的特异性和亲和力。例如，如果要检测某种特定的蛋白质（抗原），就需要使用针对该抗原的特异性抗体。

2. 固相化技术ELISA实验中，抗原或抗体通常被固定在固相载体上（如塑料微孔板）。这种固相化过程使得抗原或抗体能够稳定地吸附在微孔板表面，便于后续的反应和检测。固相化的抗原或抗体被称为“捕获相”，而加入的样本中的抗原或抗体则被称为“检测相”。

3. 酶标记技术为了实现检测信号的放大和可视化，ELISA中通常使用酶标记的抗体或抗原。常用的酶包括辣根过氧化物酶（HRP）和碱性磷酸酶（AP）。酶标记的抗体或抗原与固相化的抗原或抗体结合后，酶会与底物反应，产生可检测的信号（如颜色变化或荧光信号）。

4. 底物反应与信号检测酶与底物反应后，会产生颜色变化或荧光信号。这种信号的强度与样本中目标分子的浓度成正比。例如，辣根过氧化物酶（HRP）可以催化过氧化氢分解，使无色的底物（如TMB）变为蓝色，加入酸终止反应后变为黄色，通过分光光度计测量吸光度即可定量分析目标分子的浓度。

5. 检测模式ELISA主要有以下几种检测模式：直接ELISA：抗原固定在固相载体上，加入酶标记的抗体与抗原结合，通过底物反应检测信号。间接ELISA：抗原固定在固相载体上，先加入未标记的特异性抗体，再加入酶标记的二抗（针对一抗的抗体），通过底物反应检测信号。夹心ELISA：固相载体上固定捕获抗体，加入样本中的抗原，再加入酶标记的检测抗体，通过底物反应检测信号。夹心ELISA是检测蛋白质等大分子最常用的方法。竞争ELISA：样本中的抗原与固相载体上的抗原竞争结合酶标记的抗体，通过检测酶标记抗体的结合量来推断样本中抗原的浓度。

6. 定量分析ELISA的检测结果可以通过标准曲线进行定量分析。标准曲线是通过一系列已知浓度的标准品进行检测，绘制吸光度（或荧光强度）与浓度的关系曲线。根据样本的吸光度或荧光强度，通过标准曲线可以计算出样本中目标分子的浓度。
